The Effects of Angelica koreana Extracts on Acetamionphen-induced Hepatotoxicity


Abstract
Previous reports have shown that furanocoumarins, abundant compounds present in Angelica koreana, affect acetaminophen(AAP)-induced hepatotoxicity which is primarily associated with the formation of reactive metabolic intermediates. The present study was initiated to examine whether Angelica koreana would protect the liver from AAP-induced tissue damage. Rats were treated with each of the fractions from Angelica koreana methanol extreat at the dose of 1g/§¸, p.o., followed by the treatment with AAP(1800§·/§¸, p.o.). The fractions include hexane, ether, ethylacetate and butanol. Biochemical parameters which indicate liver damage were monitored. The rats treated with AAP alone exhibited increases in GOT, GPT and ALP activities, as compared with the saline-treated rats, whereas the serum enzyme activities from the rats treated with both Angelica koreana extracts and AAP were decreased, as compared with those from the AAP-treated rats. Hexane fraction appeared to be the most effective among the fractions examined, resulting in 111%, 76% and 88% decreases in GOT, GPT and ALP activities, respectively, relative to the treatment with AAP alone. Total cholesterol, lipid and bilirubin contents were elevated after AAP treatment. The elevations were significantly inhibited by the pretreatment with Angelica koreana extracts. Hexane fraction is the most effective, causing 85%, 126% and 89% decreases in total cholesterol, lipid and bilirubin levels as compared with the AAP-treated rats. Angelica koreana extracts failed to effect on the rate of lipid peroxidation. These results suggest that Angelica koreana extract is effective against the liver damage induced by acetaminophen and that lipid peroxidation is not involved in the protection by the plant.
